Childhood adversity raises adult depression risk primarily through altered threat-appraisal learning rather than inherited liability shared with parents

The claim is about mediation, not mere association. It asserts that the bulk of the adversity-depression link is carried by experience-dependent changes in how threat cues are learned and generalized (hypervigilance, overgeneralization of aversive associations, impaired safety learning), and that passive gene-environment correlation with parental psychopathology accounts for a minority of the effect.

This is the mechanistic leg of the claim: the sibling data show the effect is environmental, but only mediation through a threat-learning index shows it runs through threat appraisal specifically. A skeptic will note that statistical mediation with a partly-concurrent mediator cannot establish causal ordering, and that generalization gradients may be a prodromal marker of depression rather than a cause.

Sibling comparison removes shared genetic and household confounding, so it constrains how much of the association passive rGE can explain. A skeptic will attack the assumption that discordant siblings were exposed to the same family environment otherwise — non-shared confounders and differential parental treatment can inflate the residual within-pair effect.

Offers a rival mechanism that would displace threat appraisal as the primary path. The vulnerability is that the mediation model uses concurrent CRP and depression measurement, and adiposity was not modeled — BMI is a strong common cause of both CRP and depression and could generate the entire mediated path.

An experimental test of the mediator: if altered threat appraisal is the primary causal channel, moving it should move the outcome. The strongest defense is that adult CBM does not reverse a developmentally-consolidated learning history, so a null intervention effect is compatible with a real developmental mechanism — but that defense makes the claim harder to falsify.

This attacks the mediation claim at its root by showing a genetic pathway operating through the parent, which sibling designs partly absorb but do not decompose. The skeptical response is that 'genetic nurture' is still an environmental mechanism in the child — it just has a genetic upstream cause — so it may not contradict threat-appraisal mediation at all.

Suggest adding 'behavior-genetics' visibility on the child items too — half this tree is genetically-informed design work and it isn't surfacing in the field filters.
Seems obvious that trauma causes depression, not sure why this needs a whole tree.
The claim isn't that adversity is associated with depression — nobody disputes that. It's a specific mediation claim about which pathway carries the effect, and the trio and CBM-trial evidence show why that's genuinely open.
Fair, I misread the title. The MEND trial thread changed my mind about how settled the mechanism is.
Great page. Adding a note for students: notice that the sibling design and the trio design are answering different questions and both can be right. Confounding-removal and mechanism-identification are separate inferential jobs.
The word doing all the work here is 'primarily'. As written the claim needs the environmental share to exceed 50% AND the threat-appraisal share of that environmental effect to exceed the sum of all other environmental mechanisms. The evidence tree addresses the first much better than the second.
Correct, and I'd concede the second is currently underdetermined. The Okonjo disattenuated estimate is the only direct number and it has a wide CI.
Which is why rival-mechanism evidence belongs on this page rather than being treated as a distraction.
